介绍：禽流感（Avian influenza）是由A型流感病毒引起的一种人与动物共患病，不仅引起禽类以呼吸系统疾病、产蛋下降乃至急性致死和高死亡率等为特征的烈性传染病，而且可导致人的感染和死亡，严重威胁人类健康。禽流感病毒（Avian influenza virus ,AIV）显著的生物学特征之一是亚型众多，变异频繁，按病原体类型的不同，可分为高致病性、低致病性和非致病性禽流感三大类。世界卫生组织（OIE）将其列为必须通报疾病。我国2008年修订的《一、二、三类动物疫病病种明录》将其列为一类动物疫病。

用途：该试剂盒用于对AIV H5亚型进行检测，有助于对AIV H5亚型的实验室诊断、监测和流行病调查。

原理：本试剂盒采用一步法实时荧光反转录聚合酶链式反应(One-Step Real Time RT-PCR) ，结合Taqman技术，对特异核酸进行定性或定量检测。

保存期及贮藏条件：本产品-20℃避光可保存12个月。

样品收集，操作和储存

1. 选择发病早期，病毒含量比较高的咽拭子、泄殖腔拭子、血液等样本，无菌采样并立即送检。

2. 样品短期可在4℃冷藏保存，较长时间保存应在--20℃或者-70℃，并且避免反复冻融。

检测步骤

1.样本处理

取咽拭子、泄殖腔拭子上清液、血清、血浆等样本80μL于0.5mlEP管中，加入20μL样本处理液混匀备检。

备注：组织样本建议采用商品化的核酸提取试剂盒提取核酸，然后再进行后续检测。

2.扩增试剂的准备

将样品数+1份阴性对照+1份阳性对照设为N，计算试剂所需量，反应体系配制见下表

将各试剂按照表中用量加入无菌离心管中，充分混匀，短暂离心后在PCR反应管中分别加入18μL混合液。

3.加样

将制备好的样本，阴性对照，阳性对照各取2μL分别加入对应反应管中，加完后盖紧PCR反应管盖，置于PCR仪上。

注：报告基团选FAM,淬灭基团选NONE。

结果成立原则

阳性对照：CT值＜30并有明显的扩增曲线。

阴性对照：无CT值或者CT值>37，线性为直线或者轻微斜线，无明显扩增曲线。

以上要求需同时满足，否则本次实验无效，需要重新进行。

阳性，样本中检测出AIV H5亚型。

30-37

可疑，需重复检测。若第二次检测CT值仍为30-37并有明显扩增曲线则判定为阳性，否则为阴性。

>37并无扩增曲线

阴性，样本中未检测出AIV H5亚型。

注意事项

1.反应体系应在特定配液区或者超净工作台中配制，整个实验过程严格控制污染。

2.PCR整个试验分配液区、模板提取区、扩增区。流程顺序为配液区→模板提取区→扩增区。严禁器材和试剂倒流。

3.反复冻融试剂将减低检测灵敏度，本试剂盒应在5次内用完.

4.-20℃保存的各试剂管使用前应在室温下完全融化，液体融化后需彻底混匀，2000 rpm离心15 s ，使用后立即放回-20 ℃。

5.在RNA提取过程中，避免RNA酶污染，尽量缩短操作时间。

6.注意防止试剂盒组分受污染。不要使用超过有效期限的试剂，试剂盒之间的成分不要混用。

7.PCR反应管避免气泡存在，管盖需盖紧。
